In rebuiling my '67 225 with a new cam and a new chain, I cannot get the timing marks (@TDC) to meet at the centerline from crank to cam bolts. They are a good tooth off, which seems inacceptable? The crank sprocket is the oringinal, and the chain is tight. Should I be suspect of the cam sprocket or the ISKY cam? Any wisdom would be welcome.

Setting-up a cam with any accuracy requires a degree wheel or other special checks.
Search the messages for "degree cam" or "split overlap" for some ideas.

As for you "one tooth off" comment, you should be able to move the sprocket over one tooth in the chain to get closer to lined-up marks. (where-ever that puts it is a W.A.G.)

I had trouble installing a chain, I will have to take apart as I am one tooth off.
I am going to put a string the runs through the center of Cam to center of Crank to see exactly where the timing marks should line up. I have also bought a flywheel turning tool. If this fails I will go with a degree wheel.
