I haven't heard of this locknut technique, but I imagine it probably could be made to work. It would have to be a small/slim nut. Here is the valve adjustment procedure.

How accurate would this method be? I pulled the plugs, valve cover, put my thumb over the spark plug hole and cranked the engine over until it blew my thumb out of the way, then I adjusted both valves. What is the concensus on this?

I think there would be considerable room for error there. One way to accomplish what you are trying to do is the divide your balancer into 3 equal parts beginning at the TDC mark. Bring the engine to TDC on cylinder #1 compression stroke and adjust those valves then following the firing order turn the engine 1/3 of a turn and adjust the next set and continue through the firing order until they are all adjusted. You can rock the engine back and forth a few degrees to be certain that the valves are resting on the base circle of the cam. It's just as easy though to simply adjust them with the engine running then you can start at one end and work toward the other. In either case, just don't forget that the Intake/Exhaust sequence reverses between #3 and #4.

what I do is I pop the dist. cap off and turn the engine over off the starter relay when the rotor points to the #1 cyl I adjust the #1 & #6 then when it points to the #2 cyl. I adjust the #2 & #5 then the #3 & #4 to get a rough adjustment then I do the fine tunning with the engine running

Not at all accurate.

I "rough adjust" a new set-up by watching the mating valve's position and then adjusting the other.
For a final valve lash adjustment, I do that with the engine running, hot and the idle turned down as low as possible.

i have always used the old mopar race method on my stock and race engines, make a mark 180deg from tdc mark on balancer, bring engine to tdc set int #1&#4, ex#5 turn 180deg set int#5, ex#3&#6 turn 180deg set int#3&#6,ex#2 turn 180deg set int#2,ex#1&#4. do this after warmup with engine off, just play with adjustments untill you have them tight enough. 0.010 int& 0.016ex i found better with my combo :D
