I just removed the linkage and installed a throttle valve cable. what is the best way to adjust the T/V pressure?

Wide open throttle should move the lever all the way back to the kickdown position. If you achieve this and your part throttle upshifts aren't correct you can make minor adjustments, but you need to maintain kickdown. If you don't maintain enough throttle pressure you can burn up the transmission! If the part throttle shifts still aren't right then you likely have a geometry problem. Many times the cable doesn't move as far as the mechanical linkage did and so you have to move the cable attachment point on the transmission side closer to the lever's center of rotation or the throttle side farther from the center of rotation.

The mechanical linkage had slack movement prior to moving the lever on the trans. Should I have slack in the cable also with the cable just moving the lever all the way back at full throttle?

No slack should be present.
