Is it ok to prime an oil pump by putting Vaseline in the cavity and turning it by hand or do you have to take off the cover and pack it in there instead?

Vaseline in the cavity as you describe worked for me.

I would lean towards lubriplate I think it is called... (motor assembly grease) because it is oil soluble, and my understanding is that vaseline while nice and thick, it does not dissolve in the oil, but becomes bits of snot floating on the oil.

Both will work, but if I had lubriplate and vaseline on hand I would choose the assembly grease.

I have always used Vasoline. One caution, whatever you use, do not "fill" the cavity. Just enough grease to seal the rotors to each other, and the housing. If you fill the cavity, you will not be able to produce enough suction to pull oil from the pan. Don't ask how I know this.

On the Last engine I built I did not even bother with any grease or Vaseline at all, of course It was not an oil pump that had been totally disassembled. It probably had some oil left in it from the disassembly.

I Just poured oil into the 2 passages where the oil filter screws onto the pump, and kept filling those 2 ports with oil until they filled up.

Also Filled the filter 1/2 way before spinning it on.

Cranked over the engine with no Spark plugs / no coil wire and primed / built oil pressure that way. It worked fine.

I used a separate pump to prime the engine.

That said, Chrysler built millions of Slant Six engines - a few of which developed a reputation for being incredibly reliable and long lasting - and never primed any of them.

I don't think I packed the one on my Valiant before I started it.

Most of the time it is not necessary to put grease in the pump. I do it for the "one" time it might not pick up the oil. Doesn't hurt any thing, and might save some effort later.
One other thing I do at times, is no oil in engine, hook a tank, with oil to the oil gauge pressure port, with a on/off valve, open valve, pressureize the tank (fills engine passages)while turning engine by hand, close valve, start engine.
